Platelets release mitochondria serving as substrate for bactericidal group IIA-secreted phospholipase A2 to promote inflammation.
Mitochondrial DNA (mtDNA) is a highly potent inflammatory trigger and is reportedly found outside the cells in blood in various pathologies. Platelets are abundant in blood where they promote hemostasis. Although lacking a nucleus, platelets contain functional mitochondria. On activation, platelets produce extracellular vesicles known as microparticles. We hypothesized that activated platelets ...
متن کاملSimplified YM-26734 inhibitors of secreted phospholipase A2 group IIA.
Simplified analogs of YM-26734, a known inhibitor of secreted phospholipase A(2) (sPLA(2)) group IIA, were synthesized and found to also display potent inhibition at low nanomolar concentrations. Analogs were based on the didodecanoylphloroglucinol portion of YM-26734 which contains the predicted active site calcium binding group.
متن کاملGroup V Secreted Phospholipase A2 Induces the Release of Proangiogenic and Antiangiogenic Factors by Human Neutrophils
Secreted phospholipases A2 (sPLA2s) are extracellular enzymes that catalyze the release of free fatty acids and lysophospholipids from membrane phospholipids and also bind to different receptors (e.g., PLA2R1 or integrins). To date, 12 mammalian sPLA2s have been identified, which play a critical role in pathophysiological processes including inflammation and cancer. sPLA2s activate immune cells...
متن کاملHistochemical Studies on Hydrolytic Enzyme Activities
Histochemical observations were carried out on various hydrolytic enzymes in the spontaneous mammary carcinoma of C3H-strain mice. Alkaline phosphatase activity was observed in the luminal border of the tubular structure and in the periluminal surface, which showed a cystic dilatation in ducts and papillae. Capillaries in the stroma showed a high enzymatic activity. Acid phosphatase activity wa...
